Objective: 

This course will enable the students to

synthesize some organic compounds, purify them through crystallization and to inspect the purity through melting point and boiling point along with thin layer & paper chromatography.

Chemistry Laboratory III

(Practical)

 

CO33: Synthesize organic compounds and, purify them   using various solvents, including water, alcohol, and alcohol-water mixtures for crystallization.

CO34: Determine melting and boiling points of compounds using distillation and capillary methods and evaluate the effect of impurities.

CO35: Separate the components of a given mixture using paper and thin layer chromatography (TLC).

CO36: Compile and report the results efficiently, and develop communications skills.

CO37: Contribute effectively in course-specific interaction.

Approach in teaching:

Power point presentation, laboratory instructions and demonstrations.

 

 

Learning activities for the students:

Peer learning, hands-on-learning, interpretation of data, record

writing and learning through virtual labs.

 

Conduction of experiments, lab records, viva voce.

 

 

 

Unit I: 

Organic Synthesis

 

· Acetylation of aniline using green approach

· Bromination of acetanilide by conventional method/green approach

· Nitration of acetanilide by conventional method

· S-Benzylisothiouronium salt of of benzoic acid and oxalic acid.

 

 

Purification of organic compounds

· Crystallization of synthesized organic compounds using the following solvents: a. Water b. Alcohol c. Alcohol-Water      

· Determination of the melting points of above compounds and unknown organic compounds (electrically heated melting point apparatus)   

· Effect of impurities on the melting point: mixed melting point of two unknown organic compounds

· Determination of boiling point of liquid compounds. (boiling point lower than and more than 100 °C by distillation and capillary method)

 

Chromatography

Theory: Classification, principle and working of Thin Layer and Paper chromatography.

Experiments:

· Separation of a mixture of two amino acids by ascending and horizontal paper chromatography.

· Separation of a mixture of two sugars by ascending paper chromatography.

· Separation of a mixture of o-and p-nitrophenol or o-and p-aminophenol by thin layer chromatography (TLC)
