Course Objective(s):
This course will enable the students to-

explain the core concepts of purification through crystallization and to inspect the purity through melting point and boiling point along with thin layer & paper chromatography.

On completion of this course, the students will be able to-

CO28: understand the importance of purification in organic chemistry.

CO29: apply the knowledge to determine the melting and boiling points and judge the extent of purity present in the compound.

CO30: apply the concepts of ascending and horizontal paper chromatography for separation and to check the purity of organic compounds.

CO31: illustrate the concept of thin layer chromatography (TLC) for separation of compounds and analyse their purity.

CO32: develop their own solvent systems to separate a mixture of compounds.

Unit I: 
  • Checking the calibration of the thermometer 
  • Purification of organic compounds by crystallization using the following solvents: a. Water b. Alcohol c. Alcohol-Water                                    
  • Determination of the melting points of above compounds and unknown organic compounds (electrically heated melting point apparatus)                   
  • Effect of impurities on the melting point: mixed melting point of two unknown organic compounds
  • Determination of boiling point of liquid compounds. (boiling point lower than and more than 100 °C by distillation and capillary method)
     

Chromatography

Theory: Classification, principle and working of Thin Layer and Paper chromatography.

Experiments:                                                                                                    

  • Separation of a mixture of two amino acids by ascending and horizontal paper chromatography.
  • Separation of a mixture of two sugars by ascending paper chromatography.
  • Separation of a mixture of o-and p-nitrophenol or o-and p-aminophenol by thin layer chromatography (TLC)
